1. I ride a mullet bike setup - should I carry spare tubes for both wheel sizes?

No, a 27.5 tube will work fine to get you home on a 29" wheel if you are in a bind.

2. Can I put sealant inside tubes?

Yes - but check the type of sealant you want to use - some works perfect inside tubes, but will state that on the product.
